Dorsal Collection

Dorsal collection is a biological sampling approach that obtains tissue, fluid, or other specimens from the dorsal region of an organism for laboratory analysis. The procedure typically involves positioning the subject, accessing the dorsal surface with sterile instruments, and transferring the sample while limiting contamination and tissue damage. In immunology and infection research, dorsal collections can provide material for detecting pathogens, measuring immune mediators, or characterizing local host responses. Consistent sampling conditions improve reproducibility and support comparisons across experimental groups, making the method useful for studying infection progression, inflammatory responses, and treatment effects.

2017

This protocol presents the use of a dorsal root ganglion (DRG) injection with a viral vector and a concurrent dorsal root crush injury in an adult rat as a model to study sensory axon regeneration. This model is suitable for investigating the use of gene therapy to promote sensory axon regeneration.

Modeling Sensory Axon Regeneration in a Rat Using Dorsal Root Ganglion Injection and Dorsal Root Crush Injury

Source: Cheah, M., et al. Dorsal Root Ganglion Injection and Dorsal Root Crush Injury as a Model for Sensory Axon Regeneration. J. Vis. Exp. (2017).This video demonstrates a step-by-step procedure for injecting a viral suspension into the dorsal root ganglia (DRGs) and inducing a controlled axonal injury via a dorsal root crush in an anesthetized rat. The protocol involves exposing the DRGs, delivering a growth-promoting viral vector to facilitate axonal regeneration, and severing axonal...

An in vivo imaging protocol to monitor primary sensory axons following dorsal root crush is described. The procedures utilize wide-field fluorescence microscopy and thy1-YFP transgenic mice, and permit repeated imaging of axon regeneration over 4 cm in the PNS and axon interactions with the interface of the CNS.

Neuronal Enrichment of Dorsal Root Ganglia by Immunopanning

This video demonstrates a protocol for enriching neuronal cultures from dorsal root ganglia by immunopanning, thereby improving their purity and utility for neurological research.

Isolation of Macrophages from Mouse Dorsal Root Ganglion Using Mechanical Dissociation

2025

This video demonstrates an enzyme-free protocol to isolate macrophages from mouse dorsal root ganglion (DRG) using a mechanical dissociation technique. Upon homogenization of the freshly collected DRG tissue, antibodies against the cell-surface receptors on the macrophages are used to label the cells fluorescently, followed by performing flow cytometry to confirm the presence of macrophages in the sample.
