Endocytic Potential Assessment

Endocytic potential assessment measures a cell’s capacity to internalize extracellular material through endocytosis, a membrane-driven process central to nutrient uptake, receptor signaling, antigen processing, and infection. In a typical assay, cells encounter a labeled cargo such as a particle, protein, or microorganism, and internalization is quantified by fluorescence-based imaging or flow cytometry while distinguishing surface-bound material from cargo enclosed within endocytic vesicles. In immunology and infection research, these measurements help compare cellular uptake, evaluate antigen-presenting cell function, characterize host-pathogen interactions, and assess how treatments alter entry pathways. The results can clarify mechanisms of immune surveillance and microbial invasion.

Endocytic and retrograde transport of proteins from the plasma membrane to the trans-Golgi network is essential to maintaining membrane homeostasis and regulating signaling. Here, we describe a method to image and quantify endocytic transport of transmembrane cargo proteins by live-cell microscopy using derivatized anti-GFP nanobodies in HeLa cells.

Measuring Plasma Membrane Protein Endocytic Rates by Reversible Biotinylation

Regulated endocytosis governs the cell surface expression levels of the majority of membrane proteins. Here we utilize reducible, membrane impermeant biotinylation reagents to measure the endocytic rate of the dopamine transporter (DAT), a polytopic membrane protein. The method facilitates a straightforward approach to measuring the endocytic rate of most plasma membrane proteins.

Zebrafish as a Model to Assess the Teratogenic Potential of Nitrite

Exposure to teratogens may cause birth defects. Zebrafish are useful for determining the teratogenic potential of chemicals. We demonstrate the utility of zebrafish by exposing embryos to various levels of nitrite and also at different times of exposure. We show that nitrite can be toxic and cause severe developmental defects.
