Foxp3 Helios

Foxp3 and Helios are transcription factors used to characterize regulatory T cells (Tregs), immune cells that suppress excessive responses and help maintain tolerance. Foxp3 establishes the Treg transcriptional program, while Helios, an Ikaros-family DNA-binding protein, is associated with Treg development, identity, and functional stability, although it is not an exclusive marker of thymic-derived Tregs. In immunology and infection research, intracellular staining for Foxp3 and Helios, often combined with surface markers and functional assays, helps distinguish Treg populations and assess changes during immune challenge. These analyses clarify how Tregs regulate inflammation, pathogen responses, and immune-mediated tissue damage.

In this video, we describe a protocol for isolating single cells from murine pancreatic draining lymph nodes (PDLNs) and the characterization of regulatory T cells (Tregs) from the single-cell population using flow cytometry. Tregs are identified based on the positive expression of the cell-surface markers CD4, CD25, and Nrp1, along with the nuclear transcription factors Foxp3 and Helios.

Speckle tracking echocardiography is an emerging diagnostic imaging technique for the quantitative assessment of global and regional myocardial performance. Standard view echocardiographic motion images are recorded and deformation parameters are subsequently measured by automated continuous frame-by-frame tracking and motion analysis of speckles within the B-mode images of the myocardium.
