Host Protein Cleavage

Host protein cleavage is the enzymatic cutting of proteins produced by an infected organism, a process that can shape immune defense and pathogen survival. During infection, pathogen- or host-derived proteases hydrolyze specific peptide bonds, activating, inactivating, or structurally modifying targets such as receptors, complement components, cytokines, and barrier proteins. These changes can weaken epithelial protection, alter inflammatory signaling, or help microbes evade immune recognition. Studying host protein cleavage clarifies mechanisms of virulence and immune regulation while supporting the identification of infection biomarkers and the development of protease inhibitors or other therapies that preserve protective host functions.

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.
