Human Chimerism Assessment

Human chimerism assessment is the measurement of genetically distinct cell populations within one person, providing insight into cell origin, persistence, and immune status. The method compares donor- and recipient-informative genetic markers, commonly short tandem repeats or single-nucleotide polymorphisms, using quantitative PCR, digital PCR, or sequencing to estimate the proportion of each population in blood or tissue. In immunology and infection research, these measurements track engraftment and immune reconstitution after hematopoietic stem cell transplantation, help identify mixed chimerism associated with graft failure or graft-versus-host disease, and support studies of microchimerism during immune responses. Sensitive longitudinal testing can also clarify how cellular populations change during treatment or infection.
