Icam-1 Quantification

ICAM-1 quantification is the measurement of intercellular adhesion molecule 1 abundance on cell surfaces or in biological samples, providing a readout of cellular activation and inflammatory signaling. The analysis typically uses antibodies that specifically bind ICAM-1, with detection by methods such as flow cytometry, immunoassay, or immunoblotting; signal intensity or concentration is compared with standards or controls to estimate expression levels. In immunology and infection research, these measurements help assess leukocyte adhesion, endothelial activation, pathogen-induced inflammation, and responses to cytokines or therapeutic compounds. Quantifying ICAM-1 can therefore clarify immune-cell recruitment and support evaluation of disease mechanisms and treatment effects.

This article describes a method to visualize formation of an HIV-1 envelope-induced virological synapse on glass supported planar bilayers by total internal reflection fluorescence (TIRF) microscopy. The method can also be combined with immunofluorescence staining to detect activation and redistribution of signaling molecules that occur during HIV-1 envelope-induced virological synapse formation.

Studying tumor microenvironment may identify prognostic or predictive biomarkers of clinical response to immunotherapy. Presented here, is an innovative method based on in situ fluorescence multispectral imaging to analyze and count automatically various subpopulations of CD8+ T cells. This reproducible and reliable technique is suitable for large cohort analyses.
