Immunofluorescence Assay

Immunofluorescence assay is a laboratory method that uses fluorescently labeled antibodies to detect and localize specific antigens in cells, tissues, or clinical specimens. In a direct assay, a fluorophore-conjugated primary antibody binds the target; in an indirect assay, an unlabeled primary antibody is detected by a fluorescent secondary antibody, and the signal is visualized by fluorescence microscopy. In immunology and infection research, this approach reveals pathogen distribution, host immune markers, and changes in cellular structure while preserving spatial context. It supports diagnostic testing, characterization of immune responses, and investigation of host-pathogen interactions.

CTC Immunofluorescence Assay : A Technique to Characterize Circulating Tumor Cells

2023

This video describes the Circulating Tumor Cells (CTC) Immunofluorescence assay, a technique to characterize the biomarkers on circulating tumor cells in non-small cell lung cancer patient samples. This assay could be used to monitor dynamic changes of the tumor and immune response, which may reflect the response to chemotherapy, radiation, and possible immunotherapy treatments.

Immunofluorescence Assay to Detect Hepatitis C Virus Replication Complexes in Infected Cells

2026

Source: Ren, S., et al. A Protocol for Analyzing Hepatitis C Virus Replication. J. Vis. Exp. (2014).This video demonstrates an immunofluorescence-based method to detect Hepatitis C virus replication in infected hepatoma cells. The protocol involves cell fixation, antibody staining for viral protein and double-stranded RNA, and visualization using fluorophore-tagged secondary antibodies. Dual signal detection under a fluorescence microscope confirms active viral replication within the host cells.

High-Throughput Automated Multiplex Immunofluorescence Assays for Translational Research

2025

This report describes an automated protocol for multiplex immunofluorescence (mIF) assays on an automated slide stainer. It demonstrates the high reproducibility of spatial proteomics on standard high-throughput tissue autostainers and whole-slide fluorescence imagers, which is ideal for custom mIF assays across a large number of tissue slides in translational research.

Multiplexed Immunofluorescence Assay for Spatial Assessment of Senescence Markers in vivo

2026

Multiplexed immunofluorescence enables sequential labeling of multiple antibody markers within a single tissue section to support spatial analysis of tissue architecture and cell populations. Here, a workflow for multiplex detection of senescence-associated, structural, and immune markers is presented, including manual and automated slide preparation, iterative imaging, and single-cell analysis using digital pathology software.
