2-fluoro Modified Rna

2-Fluoro modified RNA is a synthetic RNA molecule in which the ribose 2′-hydroxyl group is replaced with fluorine, a change that can improve performance in biological and therapeutic settings. This substitution alters sugar conformation, strengthens some RNA duplexes, and increases resistance to nuclease degradation while potentially changing recognition by innate immune receptors. In immunology and infection research, 2′-fluoro RNA supports the design of stable aptamers, small interfering RNAs, antisense molecules, and other RNA-based probes that target pathogens or regulate host responses. These properties can extend molecular activity and improve the evaluation of RNA therapeutics in complex biological environments.

2025

This video demonstrates the application of Hybridization Chain Reaction (HCR)-Fluorescence In Situ Hybridization (FISH) for visualizing the spatial distribution of SARS-CoV-2 subgenomic RNA. Combining HCR with immunofluorescence techniques has the potential to provide insights into host-virus interactions at the single-cell level.

Studying Metabolic Brain Connectivity Using 2-Deoxy-2-[18F]Fluoro-D-Glucose Dynamic Positron Emission Tomography at the Single-subject Level

The acquisition of dynamic positron emission tomography (PET) data and reconstruction into time frames allows for metabolic brain connectivity analysis at the single-subject level. We describe a method to acquire [18F]FDG dynamic PET data of the rat brain and obtain a connectivity matrix through the extraction of time-activity curves of volumes of interest.

This article provides a detailed procedure on the solid-phase synthesis, purification, and characterization of dodecamers of RNA modified at the C2'-O-position. UV-vis and circular dichroism photometric analyses are used to quantify and characterize structural aspects, i.e., single-strands or double-strands.

Ribosomal RNA Synthesis

2020

Ribosome synthesis is a highly complex and coordinated process involving more than 200 assembly factors. The synthesis and processing of ribosomal components occurs not only in the nucleolus but also in the nucleoplasm and the cytoplasm of eukaryotic cells. Ribosome biogenesis begins with the synthesis of 5S and 45S pre-rRNAs by distinct RNA polymerases. The primary transcripts are extensively processed and modified before they are bound and folded by ribosomal proteins and assembly factors,...
