Innate T Cells

Innate T cells are T lymphocytes that combine features of adaptive T cells with the rapid, broadly reactive behavior of innate immune cells, making them important early responders to infection. Unlike conventional T cells, subsets such as natural killer T, mucosal-associated invariant T, and γδ T cells use relatively restricted receptors to recognize lipid, metabolite, or stress-associated signals presented by nonclassical molecules or expressed by infected and damaged cells. Upon activation, they rapidly release cytokines and deploy cytotoxic mechanisms that shape inflammation and support other immune cells. Studying innate T cells helps clarify host defense, immunopathology, vaccine responses, and their potential for targeted immunotherapies.

This protocol details an investigation of the early interactions between virally infected nasal epithelial cells and innate cell activation. Individual subsets of immune cells can be distinguished based on their activation in response to viral infections. They can then be further investigated to determine their effects on early antiviral responses.

Cells of the Innate Immune Response

2024

The innate immune response is an immediate and non-specific response against pathogens, acting swiftly to prevent the spread of infections. The primary cells involved in this response are phagocytes and natural killer (NK) cells. Phagocytes Phagocytes police the peripheral tissues by removing cellular debris and responding to the invasion of foreign substances or pathogens. Many phagocytes attack and remove microorganisms even before lymphocytes detect them. The human body has two general...

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.

This protocol describes a comprehensive method for assessing caspase activation (caspase-1, caspase-3, caspase-7, caspase-8, caspase-9, and caspase-11) in response to both in vitro and in vivo (in mice) models of infection, sterile insults, and cancer to determine the initiation of cell death pathways, such as pyroptosis, apoptosis, necroptosis, and PANoptosis.
