Intravital Imaging

Intravital imaging is a microscopy technique that visualizes biological processes in living animals, preserving the tissue environment and cell interactions that ex vivo methods may miss. Using fluorescent labels, specialized microscopes, and optical access to tissues, researchers track cells and molecular signals over time, often through repeated imaging of the same site. In immunology and infection research, this approach reveals how immune cells migrate, recognize targets, form contacts, and respond to invading pathogens within tissues. These observations connect cellular behavior with disease progression, host defense, and treatment response, improving understanding of dynamic processes that are difficult to capture in fixed tissue or cell culture.

Recent advances in 2-photon microscopy have enabled real-time in situ imaging of live tissues in animal models, thereby enhancing our ability to investigate cellular behavior in both physiologic and pathologic conditions. Here, we outline the preparations required to perform intravital imaging of the mouse popliteal lymph node.

We have developed novel laboratory tools and protocols for intravital imaging acquisition of the thymus. Our technique should help in the identification of “niches” within the thymus where T cell development occurs.
