Larval Homogenization

Larval homogenization is a laboratory method that mechanically disrupts whole larvae to produce a uniform tissue homogenate for downstream analysis. The process breaks open cells and tissues, typically in a suitable buffer, releasing host proteins, nucleic acids, metabolites, and microbial material into a representative sample. In immunology and infection research, researchers use larval homogenates to measure pathogen burden, detect immune-related molecules, and compare host responses under different experimental conditions. This approach supports biochemical, molecular, and microbiological assays while enabling analysis of small or delicate model organisms that are difficult to sample through conventional tissue dissection.

Test for Homogeneity

The goodness–of–fit test can be used to decide whether a population fits a given distribution, but it will not suffice to decide whether two populations follow the same unknown distribution. A different test, called the test for homogeneity, can be used to conclude whether two populations have the same distribution. To calculate the test statistic for a test for homogeneity, follow the same procedure as with the test of independence. The hypotheses for the test for homogeneity can be stated as...

Drosophila Larval IHC

Immunohistochemistry (IHC) is a technique used to visualize the presence and location of proteins within tissues. Drosophila larvae are particularly amenable to IHC because of the ease with which they can be processed for staining. Additionally, the larvae are transparent, meaning that some tissues can be visualized without the need for dissection. In IHC, proteins are ultimately detected with antibodies that specifically bind to “epitopes” within the protein of interest. In order to preserve...

Drosophila melanogaster larvae provide an ideal model system to investigate the mechanisms of axonal transport within larval segmental nerves. Using this procedure, 3rd instar larvae carrying various mutations can be compared to wild type larvae.

Larval Contact Assay: A Procedure to Evaluate Mosquito Larval Mortality following Direct Contact with Test Larvicidal Compound

In this video, we describe the larval contact assay to evaluate the toxicity of the dopamine receptor antagonist amitriptyline on the third instar stage larvae of mosquitoes, known vectors of several human infectious diseases. Assessing the larval mortality upon incubation with different amitriptyline concentrations over different time intervals helps determine its lethal concentration 50.
