Macrophage Interaction

Macrophage interaction refers to the ways macrophages communicate with pathogens, neighboring immune cells, and tissue cells to coordinate host defense and tissue responses. These cells detect microbial molecules through pattern-recognition receptors, engulf targets by phagocytosis, and release cytokines and chemokines that recruit and activate other leukocytes. Macrophages can also present antigens to T cells, linking innate and adaptive immunity, while pathogens may alter macrophage signaling or survival to evade clearance. Studying these interactions clarifies how infections are controlled, how excessive inflammation causes tissue damage, and how therapies might improve antimicrobial immunity or limit immunopathology.

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.
