Monocyte Enrichment

Monocyte enrichment is a cell-separation process that increases the proportion of monocytes in a mixed leukocyte sample for downstream analysis or experimentation. It commonly uses density-gradient centrifugation to recover peripheral blood mononuclear cells, followed by antibody-based magnetic selection or depletion, in which labeled cells bind magnetic beads and are retained or removed in a magnetic field. Enriched monocytes support studies of innate immune signaling, phagocytosis, cytokine production, differentiation into macrophages or dendritic cells, and responses to infectious agents. Consistent enrichment improves sample comparability while preserving cells for functional assays, molecular profiling, and host-pathogen research.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

This manuscript describes the procedure of real time imaging of the mouse calvaria bone marrow to study the monocyte mobilisation mechanism.
