Murine Lung Model

Murine lung models are experimental systems that use mice to study respiratory biology, immune responses, and infectious disease in a living organism. Researchers introduce pathogens, antigens, or inflammatory stimuli into the airways and assess how epithelial barriers, alveolar macrophages, neutrophils, lymphocytes, and cytokine signaling shape pulmonary inflammation and pathogen clearance. These models can reproduce selected features of pneumonia, viral infection, tuberculosis, asthma, and acute lung injury, while enabling controlled sampling of lung tissue, bronchoalveolar lavage fluid, and blood. Findings help clarify host-pathogen interactions, evaluate vaccines and therapeutics, and identify mechanisms that may guide human respiratory disease research.

Development of Obliterative Bronchiolitis in a Murine Model of Orthotopic Lung Transplantation

Obliterative bronchiolitis is the key impediment to the long-term survival of lung transplant recipients and the lack of a robust preclinical model precludes examining obliterative bronchiolitis immunopathogenesis. Unlike other solid organ transplants, vascularized mouse lung transplantation has only recently been developed. Here we show our independently developed obliterative bronchiolitis model after murine orthotopic single-lung transplantation.

This video demonstrates a method of producing decellularized extracellular matrix or ECM scaffolds by applying antegrade perfusion of detergents through the vasculature. The structural ECM scaffolds can then be used to analyze ECM remodeling associated with cardiopulmonary disease.

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.

An In Vitro Method to Identify Innate and Adaptive Immune Cells Residing in Murine Lungs

2025

This video demonstrates an in vitro technique to identify immune cells residing in murine lungs. After isolating target immune cells from murine lung tissue, they are surface-labeled using a cocktail of fluorophore-tagged antibodies against specific markers on innate and adaptive immune cells. Additionally, monocytes, macrophages, and dendritic cells are labeled using an intracellular marker. The various immune cell types are then identified using flow cytometry.
