Nadh Dehydrogenase Assay

The NADH dehydrogenase assay measures the activity of enzymes that initiate electron transfer by oxidizing NADH to NAD+, making it a useful indicator of cellular respiratory metabolism. In a typical assay, NADH oxidation is monitored spectrophotometrically through the decrease in absorbance at 340 nm, often in the presence of an electron acceptor or membrane-associated enzyme preparation. In immunology and infection research, the assay helps evaluate mitochondrial or microbial respiratory function, compare metabolic responses during infection, and assess how antimicrobial compounds or host conditions affect electron transport. These measurements can clarify changes in pathogen viability, energy production, and host-cell bioenergetics.

Mitochondria contain several flavin-dependent enzymes that can produce reactive oxygen species (ROS). Monitoring ROS release from individual sites in mitochondria is challenging due to unwanted side reactions. We present an easy, inexpensive method for direct assessment of native rates for ROS release using purified flavoenzymes and microplate fluorometry.

NADH Fluorescence Imaging of Isolated Biventricular Working Rabbit Hearts

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.
