Opsonized Erythrocytes

Opsonized erythrocytes are red blood cells coated with antibodies, complement fragments, or other opsonins that mark them for immune recognition and removal. In immunology and infection, opsonization promotes binding to Fc or complement receptors on phagocytes, allowing macrophages in the spleen and liver to engulf and degrade the labeled cells. This process provides a model for studying antibody and complement activity, receptor-mediated clearance, and defects that contribute to immune-mediated hemolysis. Laboratory assays using opsonized erythrocytes can measure serum opsonic capacity and evaluate pathways involved in host defense and inflammatory disease.

This video demonstrates an assay to monitor independent macrophage-mediated red blood cell (RBC) phagocytosis events. Mouse immunoglobulin G (IgG)-opsonized human RBCs are incubated with mouse macrophages and observed under a confocal microscope. The RBCs are engulfed by macrophages via a phagocytic cup formation and targeted for lysosomal degradation.
