Pathogen Identification

Pathogen identification is the process of determining which microorganism or infectious agent is responsible for disease, an essential step in diagnosis, treatment, and infection control. It combines analysis of clinical specimens with methods such as microscopy, culture, antigen detection, serology, and nucleic acid amplification, which recognize characteristic cellular structures, immune responses, or genetic sequences. In immunology and infection research, these approaches distinguish bacteria, viruses, fungi, and parasites while clarifying how host defenses respond to infection. Accurate identification supports targeted antimicrobial selection, outbreak tracking, surveillance of emerging pathogens, and development of vaccines and diagnostic tools.

Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) and molecular techniques (16S rRNA gene sequencing) permit the identification of rare bacterial pathogens in routine diagnostics. The goal of this protocol lies in the combination of both techniques which leads to more accurate and reliable data.

Here we present a protocol for obtaining entomopathogenic fungi from a forest wood borer and a substitutive way to evaluate their entomopathogenic activities using a Coleopteran model insect. Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.
