Peritoneal Massage

Peritoneal massage is a gentle abdominal manipulation used to distribute fluid and collect material from the peritoneal cavity, making it useful in immunology and infection research. After intraperitoneal instillation of a lavage solution, controlled massage helps mix the fluid with peritoneal surfaces and dislodge resident or recruited cells, microorganisms, and soluble mediators. Researchers can then recover the lavage for cytology, immune-cell profiling, cytokine measurement, pathogen detection, or other analyses. Standardized pressure, duration, and animal handling are important because inconsistent massage can alter sample yield and composition, affecting comparisons between experimental groups.

The peritoneal cavity in mammals contains different immune cell populations crucial for innate immune responses. An efficient isolation method is required for biochemical and functional analyses of these cells. Here we provide a comprehensive method for the isolation of peritoneal cavity cells in the mouse.
