Viral Surface Glycoproteins

Viral surface glycoproteins are carbohydrate-modified proteins displayed on the envelopes or capsids of many viruses, where they control interactions with host cells and shape immune recognition. They bind specific receptors on susceptible cells and, in enveloped viruses, can undergo structural changes that promote membrane fusion and genome entry; their exposed regions also serve as targets for antibodies. In immunology and infection research, studying these proteins helps explain host range, tissue tropism, transmission, and antigenic variation. Their properties guide vaccine design, diagnostic assay development, antiviral antibody discovery, and surveillance of emerging viral strains.

Adenovirus particles are engineered to contain either the unnatural amino acid analogue azidohomoalanine or the azido sugar O-GlcNAz. The azide group of each is chemoselectively ligated via "click" chemistry reactions as a means of viral surface modification.

African trypanosomes grown in vitro undergo antigenic variation at a low rate, such that populations are made up of parasites expressing a dominant variant surface glycoprotein (VSG) type and a small population of "switched" variants. This protocol describes a fast method for detecting and quantifying these populations.

This video demonstrates a viral attachment assay for screening antiviral compounds. Using genetically modified hepatitis C virus and luciferase, it assesses inhibitory effects on viral attachment, as indicated by lower light intensity in co-treated wells compared to wells treated with the virus alone.
