Virus Infection Assay

A virus infection assay is a laboratory method used to detect, measure, or characterize viral infection in cells, tissues, or biological samples, providing essential evidence for immunology and infection research. Depending on the assay design, researchers expose susceptible cells to a virus and quantify infection through viral nucleic acids, proteins, reporter signals, cytopathic effects, or infectious particle production. These measurements can reveal viral replication, host-cell responses, and neutralizing antibody activity. Virus infection assays support vaccine and antiviral evaluation, diagnostic development, pathogenesis studies, and analysis of how immune defenses limit or promote infection.

An Assay to Determine Polymicrobial Biofilm Initiation on Virus-Infected Cells

2025

This video illustrates the initiation of polymicrobial biofilm formation on virus-infected cells by bacteria and fungi. Herpes simplex virus infection impedes bacterial attachment to the cells, preventing bacterial-fungal co-localization. In contrast, uninfected cells display bacterial-fungal co-localization and initiation of biofilm formation.

Impedance-Based Cellular Assay to Measure Multiplicity of Infection of Influenza Virus

2025

In this video, we discuss the correlation between the CIT50 and the multiplicity of viral infections. The virus-infected cells display cytopathic effects, causing their death and detachment from the gold microelectrode, which in turn, reduces the impedance.

A Focus-Forming Assay for Quantifying Virus Titers in the Organs of an Infected Mouse

2025

The video demonstrates a procedure for quantifying the Zika virus in multiple mouse organs via a focus-forming assay. The process involves infecting mammalian cell monolayers with virus-infected tissue homogenates, followed by the use of methyl cellulose to prevent viral spread. Subsequently, immunostaining is employed to enable precise quantification and measurement of organ-specific infectivity.

Virus Plaque Assay: A Technique to Measure Infectious Herpes Simplex Virus via Quantification of Plaques Formed in Monolayer of Virus-Infected Cells

2025

This video describes the virus plaque assay of the infectious herpes simplex virus or HSV. The diluted viral particles are inoculated to the monolayer of epithelial cells. Upon incubation, the virus forms plaques and is counted visually to determine the infectious virus titer.

Influenza A Virus Studies in a Mouse Model of Infection

Influenza A viruses (IAVs) are important human respiratory pathogens. To understand the pathogenicity of IAVs and to perform preclinical testing of novel vaccine approaches, animal models mimicking human physiology are required. Here, we describe techniques to evaluate IAV pathogenesis, humoral responses and vaccine efficacy using a mouse model of infection.
