Bogota Bag

A Bogota bag is a temporary abdominal closure technique that protects exposed viscera when surgeons cannot safely close the abdomen immediately. The method uses a sterile, flexible plastic sheet, often fashioned from an irrigation or intravenous fluid bag, which is secured to the abdominal wall to cover the organs while accommodating edema and increased intra-abdominal volume. In medicine, this open-abdomen approach can help reduce tension during critical care, facilitate planned reoperation, and allow monitoring of abdominal contents after trauma, infection, or major surgery. Definitive closure is performed once swelling and the patient’s condition improve.

We present a simple and efficient protocol for the generation of human macrophages. Buffy coats are processed by double density gradient centrifugation and isolated monocytes are then differentiated to macrophages in Teflon-coated cell culture bags. This maximizes macrophage yields and facilitates cell harvesting for subsequent experiments.
