Calcium Sensitivity

Calcium sensitivity is the degree to which a cell or tissue responds to a given concentration of calcium, a concept central to muscle contraction and physiological regulation. In muscle, calcium activates contractile proteins by binding troponin C in skeletal and cardiac muscle or calmodulin-linked pathways in smooth muscle; sensitivity changes when these signaling and protein interactions alter the force produced at the same calcium level. In medicine, calcium sensitivity helps explain differences in cardiac performance, vascular tone, and skeletal muscle function, while measurements of calcium-responsive contraction support research into heart failure, hypertension, neuromuscular disorders, and treatments that modify contractility.

With its small transparent body, well-documented neuroanatomy and a host of amenable genetic techniques and reagents, C. elegans makes an ideal model organism for in vivo neuronal imaging using relatively simple, low-cost techniques. Here we describe single neuron imaging within intact adult animals using genetically encoded fluorescent calcium indicators.
