Cell Integration Analysis

Cell integration analysis is the study of how introduced, transplanted, or engineered cells become established within host tissues and contribute to tissue structure or function. It combines methods such as microscopy, lineage tracing, molecular profiling, and functional assays to determine cell survival, localization, differentiation, communication, and connectivity with surrounding cells. In medicine, researchers use these measurements to assess cell-based therapies, tissue engineering, organoid models, and regenerative strategies while distinguishing true integration from temporary cell retention. This evidence helps evaluate treatment safety and effectiveness, clarify tissue-repair mechanisms, and guide the design of more reliable regenerative interventions.

Cell-Free DNA Integrity Analysis in Urine Samples

A method for analyzing DNA integrity in the cell-free supernatant fraction of urine samples is proposed. The method is suitable for early detection of urological malignancies and has proven accurate for the early diagnosis of bladder cancer.

This manuscript describes the experimental procedure and software analysis for a bidirectional integration site assay that can simultaneously analyze upstream and downstream vector-host junction DNA. Bidirectional PCR products can be used for any downstream sequencing platform. The resulting data are useful for a high-throughput, quantitative comparison of integrated DNA targets.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.

In this study, we describe a straightforward method to perform Evans Blue Dye (EBD) analysis on zebrafish larvae. This technique is a powerful tool for the characterization of skeletal muscle integrity and delineation of zebrafish models of muscular dystrophy, and is a valuable method for the development of novel therapeutics.
