Epicardial Mapping

Epicardial mapping is a cardiac electrophysiology technique that records electrical activity from the heart’s outer surface to identify abnormal impulse formation and conduction. During the procedure, clinicians place electrodes on the epicardium, often through a surgical or percutaneous approach, and analyze activation timing, voltage, and conduction patterns to create a spatial map of cardiac tissue. These maps help distinguish scarred myocardium from viable tissue and localize arrhythmia sources, including ventricular tachycardia, when endocardial recordings are insufficient. In medicine, epicardial mapping supports individualized ablation planning, improves understanding of cardiac conduction disorders, and can guide treatment of complex rhythm disturbances.

Here, we describe methods for isolating primary mouse epicardial cells by an outgrowth culture assay and assessing the functional migration of epicardial-derived cells (EPDC) using an ex vivo heart culture system. These protocols are suitable for identifying genetic and chemical modulators of epicardial epithelial-to-mesenchymal transition (EMT) and motility.

The epicardium is an essential source of multipotent cardiovascular progenitor cells and paracrine factors that are required for cardiovascular development and regeneration. We describe here a method to culture mouse embryonic epicardial cells.
