Functional Lung Mri

Functional lung MRI is a group of magnetic resonance imaging techniques that evaluate pulmonary function, including ventilation, perfusion, and gas exchange, rather than showing anatomy alone. Depending on the method, inhaled hyperpolarized helium-3 or xenon-129 produces an MRI signal that maps regional gas distribution, while oxygen-enhanced MRI detects changes in tissue relaxation as oxygen moves through the lungs. These measurements can reveal uneven ventilation, impaired gas transfer, or altered pulmonary blood flow in conditions such as asthma, chronic obstructive pulmonary disease, and cystic fibrosis. Because MRI uses no ionizing radiation, it supports repeated assessment of disease progression and treatment response.

This protocol describes the steps and data analysis required to successfully perform optogenetic functional magnetic resonance imaging (ofMRI). ofMRI is a novel technique that combines high-field fMRI readout with optogenetic stimulation, allowing for cell type-specific mapping of functional neural circuits and their dynamics across the whole living brain.

Phase-Resolved Functional Lung MRI for Pulmonary Ventilation and Perfusion (V/Q) Assessment

Here, we describe the implementation of phase-resolved functional lung MRI as a contrast-agent-free proton MR technique for the assessment of pulmonary ventilation and perfusion dynamics. Validated and applicable across different field strengths and age groups, it could enhance clinical decision-making in the future by aiding in disease quantification and therapy monitoring.

Functional Neuroimaging Using Ultrasonic Blood-brain Barrier Disruption and Manganese-enhanced MRI

A technique is described for broadly opening the blood-brain barrier in the mouse using microbubbles and ultrasound. Using this technique, manganese can be administered to the mouse brain. Because manganese is an MRI contrast agent that accumulates in depolarized neurons, this approach enables imaging of neuronal activity.
