Fungiform Papillae Collection

Fungiform papillae collection is a tissue-sampling procedure used to obtain the mushroom-shaped structures on the anterior tongue that contain taste buds and associated sensory cells. The process involves identifying these papillae on the lingual surface, carefully excising or isolating them, and preserving the tissue for microscopic, histological, or molecular analysis. In medicine and sensory biology, collected papillae support studies of taste perception, epithelial maintenance, nerve interactions, and tissue regeneration. This approach can also help investigators examine how disease, medications, aging, or injury alter taste function and the cellular organization of the tongue.

Knowledge of molecular mechanisms underlying gustatory transduction has recently enjoyed significant advances, largely due to using animal models. However, the wide diversity in taste sensitivity and specificity among mammals warrants studies in human tissue. We describe a biopsy technique to collect living taste cells from the papillae on human tongue.

We aimed to develop a reproducible protocol for isolating and maintaining long-term cultures of human fungiform taste papillae cells. Cells from human fungiform papillae obtained by biopsy were successfully maintained in culture for more than eight passages (12 months) without loss of viability.
