Fxa Generation Assay

The Fxa Generation Assay is a laboratory method that measures the formation of factor Xa, a key enzyme in the blood-coagulation cascade and a major target of anticoagulant therapy. In the assay, coagulation is initiated in a plasma or purified protein sample, and generated factor Xa cleaves a specific chromogenic or fluorogenic substrate, producing a signal proportional to enzymatic activity. By quantifying factor Xa generation, researchers can assess coagulation potential, characterize the effects of factor Xa inhibitors, and investigate abnormalities in hemostasis. The method supports anticoagulant development, medication monitoring, and mechanistic studies of thrombin formation in medicine.

The ATP Bioluminescence Assay

2023

In fireflies, the luciferase enzyme converts a compound called luciferin into oxyluciferin, and produces light or “luminescence” as a result. This reaction requires energy derived from ATP in order to proceed, so researchers have exploited the luciferase-luciferin interaction to gauge ATP levels in cells. Given ATP’s role as the cell’s currency of energy, the ATP bioluminescence assay can provide insight into cellular metabolism and overall cell health. In this video, JoVE discusses cellular...
