Immunosuppressant Treatment

Immunosuppressant treatment uses medicines to reduce or regulate immune activity when immune responses threaten transplanted tissues or damage the body in autoimmune and inflammatory diseases. These drugs act through distinct mechanisms: calcineurin inhibitors reduce interleukin-2 production and T-cell activation, corticosteroids suppress inflammatory gene expression, and antimetabolites limit lymphocyte proliferation. Biologic agents can further target specific cytokines, receptors, or immune-cell populations, allowing more selective control of disease pathways. In medicine, treatment supports organ transplantation and manages conditions such as rheumatoid arthritis and inflammatory bowel disease, while careful dosing and monitoring help balance therapeutic benefit against infection, toxicity, and other complications.

Here we describe a high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) assay to quantify the immunosuppressant tacrolimus in dried blood spots using a simple manual protein precipitation step and online column extraction.

Establishment of A Mouse Sepsis-Induced Immunosuppression Model By Intranasal Instillation of Bacteria

2026

Here, we present a protocol for establishing a mouse sepsis immunosuppression model via intranasal instillation of Klebsiella pneumoniae and validated the model through various indicators.

Methods to Evaluate Cytotoxicity and Immunosuppression of Combustible Tobacco Product Preparations

Using optimized human peripheral blood mononuclear cell (PBMC) ex vivo assays, we showed that a combustible tobacco product preparation markedly suppresses receptor-mediated intracellularly secreted cytokines and cytolytic ability of effector PBMCs. These rapid assays may be useful in product evaluation and understanding the potential long-term effects of tobacco exposure.

Psoriasis is a chronic, immune-mediated inflammatory skin disease. The Goeckerman regimen, formulated for the treatment of psoriasis, consists of exposure to ultraviolet B (UVB) light and application of crude coal tar (CCT). The following protocol is for the administration of Goeckerman therapy for the treatment of moderate-to-severe psoriasis.

This protocol describes techniques to measure infectious outcomes underlying secondary hospital-acquired infections in the immunosuppressive condition, first by establishing cecal ligation/puncture mice then challenging them with intranasal infection to create a clinically relevant model of immunosuppression sepsis.
