Lps Induction

LPS induction is the experimental activation of an inflammatory or immune response using lipopolysaccharide, a component of the outer membrane of Gram-negative bacteria. LPS binds the CD14–MD-2–TLR4 receptor complex on immune cells, triggering intracellular signaling through NF-κB and related pathways that increase cytokine production and other inflammatory mediators. In medicine, this approach helps researchers model innate immune activation, infection-associated inflammation, and aspects of sepsis in cells or animal systems. These models support investigation of disease mechanisms, evaluation of anti-inflammatory treatments, and assessment of how immune responses contribute to tissue injury and clinical outcomes.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.
