Lymphocyte Count

Lymphocyte count is the measured number of lymphocytes in a blood sample, providing information about immune system activity and overall health. Usually reported as part of a complete blood count with differential, it is determined by analyzing circulating white blood cells and distinguishing lymphocytes from neutrophils, monocytes, eosinophils, and basophils. In medicine, an unusually high or low count can support evaluation of infections, immune disorders, inflammatory conditions, and some blood cancers, although results must be interpreted alongside symptoms, age, medications, and other laboratory findings. Repeated measurements can also help monitor disease progression or response to treatment.

Using a Hemacytometer to Count Cells

2023

Many biomedical experiments require manipulation of a known quantity of cells, in order to achieve accurate, reproducible, and statistically-relevant data. Therefore, learning how to count cells is a particularly essential technique for any successful biomedical scientist. The most common way to count cells is by using a hemacytometer - an instrument that bears two laser-etched grids, which aid in the enumeration of an aliquot cells under a simple light microscope. This data can then be used to...

In this video, we demonstrate the isolation of lymphocytes from a harvested mouse Peyer's patch. The isolated lymphocytes can be used for further analysis.

An In Vitro Technique to Stimulate Lymphocytes via Pathogenic Bacteria

2025

This video demonstrates an in vitro technique to stimulate lymphocytes via non-typeable Haemophilus influenzae (NTHi) and to assess the immune response. Incubating isolated peripheral blood mononuclear cells (PBMCs) with NTHi causes the bacterial cells to stimulate T lymphocytes, leading to cytokine production. Upon inhibiting the secretion of cytokines, immunofluorescence staining is performed to detect the accumulated intracellular cytokines in stimulated T lymphocytes.

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.
