Mammary Architecture

Mammary architecture is the organized arrangement of epithelial ducts, lobules, connective tissue, blood vessels, and surrounding stroma that enables the mammary gland to develop and function. Its structure changes through branching morphogenesis and hormone-dependent remodeling, with epithelial cells forming ductal and alveolar units supported by interactions with the extracellular matrix and stromal cells. In medicine, analyzing mammary architecture helps explain normal development, lactation, tissue regeneration, and the progression of diseases such as breast cancer. Histology, imaging, and three-dimensional culture models use these structural features to assess tissue organization, identify abnormalities, and improve diagnosis and therapeutic research.

Transfer of Mammary Gland-forming Ability Between Mammary Basal Epithelial Cells and Mammary Luminal Cells via Extracellular Vesicles/Exosomes

This protocol describes methods for purifying, quantitating, and characterizing extracellular vesicles (EVs)/exosomes from non-adherent/mesenchymal mammary epithelial cells and for using them to transfer mammary gland-forming ability to luminal mammary epithelial cells. EVs/exosomes derived from stem-like mammary epithelial cells can transfer this cell property to cells that ingest the EVs/exosomes.

Sectioning Mammary Gland Whole Mounts for Lesion Identification

We developed a method to successfully remove, process, section, and stain, for histopathological evaluation, mammary tissue that had originally been fixed on slides as whole mounts. This method may promote the collection and evaluation of mammary gland whole mounts in reproductive and developmental test guideline studies.

Intraductal Delivery to the Rabbit Mammary Gland

Here, we describe a technique for the localized delivery of reagents to the rabbit mammary gland via an intraductal injection. In addition, we describe a protocol for visualization and the confirmation of delivery by high-resolution ultrasound imaging of contrast agents.

This video describes a technique of deriving extracellular matrix hydrogels by decellularizing murine mammary fat pads following ex vivo irradiation for in vitro studies. This hydrogel helps to mimic the in vivo breast tissue environment post-radiation treatment to study tumor cell behavior.
