Mouse Artery Puncture

Mouse artery puncture is a laboratory technique used to access an artery in a mouse for collecting arterial blood or obtaining physiological measurements. The procedure involves carefully inserting a sterile needle or catheter into an exposed artery, allowing blood to be withdrawn or pressure and blood gases to be assessed while minimizing tissue trauma and maintaining hemostasis afterward. In medicine and biomedical research, arterial sampling supports studies of cardiovascular function, oxygenation, acid–base balance, pharmacokinetics, and disease models. Proper anesthesia, aseptic technique, anatomical knowledge, and monitoring are essential for reliable results and animal welfare.

We demonstrate in the video a method for producing a middle cerebral artery occlusion in adult mice using an intraluminal monofilament. We also show how to evaluate the extent of cerebral infarction by 2,3,5-triphenyltetrazolium chloride (TTC) staining.
