Pancreatic Graft Preparation

Pancreatic graft preparation is the surgical and preservation process that converts a donor pancreas into a viable organ for transplantation, a critical step in restoring endocrine function for selected patients with severe diabetes. After procurement, the graft is cooled in preservation solution, carefully inspected and dissected, and its vessels are reconstructed while preserving the pancreatic duct and an attached duodenal segment for drainage. Proper preparation limits ischemic and mechanical injury, supports safe vascular anastomosis, and helps maintain insulin-producing tissue during implantation. In clinical medicine, standardized graft preparation improves transplant quality, reduces complications, and contributes to durable glycemic control in recipients.

A plasma-induced polymerization procedure is described for the surface-initiated polymerization on polymer membranes. Further postmodification of the grafted polymer with photochromic substances is presented with a protocol of conducting permeability measurements of light-responsive membranes.

We describe a reproducible method of preparing mouse pancreatic acinar cells from a mouse for the purpose of examining acinar cell calcium signals and cellular injury with physiologically and pathologically relevant stimuli. A method for adenoviral infection of these cells is also provided.
