Photochemical Injury Model

A photochemical injury model is an experimental method that uses light-activated chemical compounds to produce controlled tissue damage, allowing researchers to study injury and repair in a reproducible setting. After a photosensitizer accumulates in the target tissue, exposure to a defined wavelength of light generates reactive oxygen species that trigger oxidative stress, cellular injury, inflammation, and structural changes. In medicine, this model can reproduce selected features of conditions such as vascular or retinal damage and supports investigation of disease mechanisms, imaging findings, therapeutic targets, and treatment efficacy. Its controllable timing and severity also make it useful for comparing interventions and studying tissue recovery.

The goal of this protocol is to photochemically induce ischemic injury to the posterior optic nerve in rat. This model is critical to studies of the pathophysiology of posterior ischemic optic neuropathy, and therapeutic approaches for this and other optic neuropathies, as well as of other CNS ischemic diseases.

Athletes absorb several hundred mild traumatic brain injuries (mTBI)/concussions every year; however, the consequence of these on the brain is poorly understood. Therefore, an animal model of single and repetitive mTBI that consistently replicates clinically relevant symptoms provides the means to advance the study of mTBI and concussion.

Lateral Fluid Percussion: Model of Traumatic Brain Injury in Mice

Lateral fluid percussion (LFP), an established model of traumatic brain injury in mice, is demonstrated. LFP fulfills three major criteria for animal models: validity, reliability and clinical relevance. The procedure, consisting of surgical craniotomy, fixation of hub followed by induction of injury, resulting in focal and diffuse injuries, is described.
