Spheroid Culture Models

Spheroid culture models are three-dimensional cell clusters grown in vitro to reproduce key features of tissue architecture, making them valuable for studying human biology and disease. Cells aggregate and self-organize under non-adherent or low-attachment conditions, creating spatial gradients of oxygen, nutrients, and signaling molecules that produce distinct proliferative, quiescent, hypoxic, and sometimes necrotic regions. In medicine, spheroids model tumor growth, drug penetration, treatment response, and cell-cell interactions more realistically than many two-dimensional cultures. They support cancer research, toxicity testing, and the development of personalized therapies while providing a controllable platform for investigating tissue behavior.

This video demonstrates a method to establish a three-dimensional tumor spheroid and T cell co-culture. An agarose cast with microwells containing tumor spheroids is prepared, followed by the addition of T cells. After incubation, this setup allows studying the interaction of T cells with tumor spheroids in a controlled three-dimensional environment.
