Biosafety Level One

Biosafety Level One (BSL-1) is the foundational laboratory containment level for work with biological materials that pose minimal risk to healthy adults, making it important for routine teaching and research. It relies on standard microbiological practices, including hand hygiene, appropriate personal protective equipment, surface decontamination, and proper waste disposal, while permitting open-bench procedures when no aerosol or higher-risk hazard is present. In neuroscience, BSL-1 facilities support studies using nonpathogenic microorganisms, established cell lines, and other low-risk materials to investigate neural development, cellular function, and experimental methods while maintaining essential laboratory safety.

Work with infectious Ebola viruses is restricted to biosafety level 4 laboratories. Tetracistronic minigenome-containing replication and transcription-competent virus like particles (trVLPs) represent a lifecycle modeling system that allows us to safely model multiple infectious cycles under biosafety level 2 conditions, relying exclusively on Ebola virus components.

Here, we present a protocol to generate pseudotyped particles in a BSL-2 setting incorporating the spike protein of the highly pathogenic viruses Middle East respiratory syndrome and severe acute respiratory syndrome coronaviruses. These pseudotyped particles contain a luciferase reporter gene allowing quantification of virus entry into target host cells.
