Caudal Ganglionic Eminence

The caudal ganglionic eminence (CGE) is a transient embryonic region of the ventral telencephalon that produces specialized neurons essential for forebrain development. During neurogenesis, progenitor cells in the CGE generate predominantly late-born inhibitory interneurons, which migrate tangentially into the developing cerebral cortex and hippocampus before differentiating into distinct neuronal subtypes. Studying the CGE helps explain how neuronal diversity, migration, and circuit organization arise during development. Its molecular programs and developmental trajectories also provide a framework for investigating neurodevelopmental disorders, modeling human brain formation, and improving strategies for generating specific interneuron populations from stem cells.

Viral-Mediated Labeling and Transplantation of Medial Ganglionic Eminence Cells for In Vivo Studies

2025

This video demonstrates the viral labeling and transplantation of medial ganglionic eminence (MGE) cells for in vivo studies. It outlines the steps involved in transducing MGE cells with a viral reporter, transplanting these transduced cells into the cortex of a mouse pup, and enabling reporter expression in specific interneuron subtypes.

GABAergic cortical interneuron progenitors disperse, develop and synaptically integrate into a host cortex after transplantation. These cells can be easily transduced before transplantation for in vivo studies of genetically modified GABAergic precursors. Here, we show viral labeling techniques to target specific interneuron subgroups using existing Cre lines and Cre-dependent reporters.
