Cerebellar Stem Cells

Cerebellar stem cells are neural stem or progenitor cells that generate cerebellar cell types and help researchers understand how this brain region develops and functions. Through self-renewal and regulated division, they produce progeny that differentiate into neurons and glial cells in response to developmental signals, tissue environment, and lineage-specific cues. Studying these cells clarifies cerebellar development, circuit formation, and responses to injury, while cultured cells and cerebellar organoids support disease modeling and drug research. Their properties may also inform future approaches to neural repair, although effective therapeutic use requires precise control of cell identity, maturation, and integration.

A Technique to Generate a 2D Monolayer of Cerebellar Cells from Induced Pluripotent Stem Cells

2025

This video demonstrates a protocol for differentiating human induced pluripotent stem cells into cerebellar progenitors. It involves transferring stem cell colonies to an ultra-low attachment plate to form embryoid bodies, promoting differentiation with growth factors, and then transferring the embryoid bodies onto a matrix-coated plate for adhesion. Subsequent steps involve inducing cell migration and adding a maturation medium to guide the development of glutamatergic and GABAergic cerebellar...

Generating Mature Cerebellar Organoids From Induced Pluripotent Stem Cells Using Single-Use Bioreactors

2025

This video demonstrates a technique for generating mature cerebellar organoids from induced pluripotent stem cells (iPSCs) using a single-use 3D bioreactor. The bioreactor facilitates iPSC aggregate formation, which then undergoes treatment with a series of culture media enriched in growth factors and chemokines that induce the differentiation of the iPSC aggregates into mature cerebellar organoids. Subsequently, the organoids are harvested and cryopreserved for further analysis.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Analyzing Purkinje Cell Survival in Organotypic Cerebellar Slice Cultures

This video illustrates a technique for obtaining cerebellar organotypic slices from a mouse pup's brain and culturing the slices in media supplemented with nutrients and growth factors to mimic cerebellar development in vitro. Additionally, specific cerebellar slices are treated with a test compound, and the compound's neuroprotective ability is analyzed using an immunofluorescence staining technique.
