Controlled Axonal Injury Model

Controlled axonal injury model is an experimental system that produces a reproducible lesion in axons so researchers can study neuronal damage and repair. By severing axons at a defined location and time, often under live-cell imaging, the model enables measurement of Wallerian degeneration, distal axon fragmentation, and subsequent regeneration or failure. In neuroscience, it supports analysis of injury responses, including cytoskeletal breakdown, calcium-dependent signaling, and interactions between neurons and glial cells. Its controlled design improves comparisons among genetic backgrounds, treatments, and experimental conditions, helping evaluate mechanisms and candidate therapies for axonal repair.

Source: Cheah, M., et al. Dorsal Root Ganglion Injection and Dorsal Root Crush Injury as a Model for Sensory Axon Regeneration. J. Vis. Exp. (2017).This video demonstrates a step-by-step procedure for injecting a viral suspension into the dorsal root ganglia (DRGs) and inducing a controlled axonal injury via a dorsal root crush in an anesthetized rat. The protocol involves exposing the DRGs, delivering a growth-promoting viral vector to facilitate axonal regeneration, and severing axonal...

Axonal Regeneration in the Sciatic Nerve of a Mouse Following a Crush Injury

Source: Li, Q., et al. Investigating Mammalian Axon Regeneration: In Vivo Electroporation of Adult Mouse Dorsal Root Ganglion. J. Vis. Exp. (2018)This video demonstrates the process of axonal regeneration in a mouse sciatic nerve following a crush injury. The isolated nerve is fixed and prepared for imaging, with fluorescent proteins expressed in neurons to visualize axon growth. Using an epifluorescence microscope, axon regeneration is traced from the injury site to assess the extent of...
