Courtship Behavior Analysis

Courtship behavior analysis is the systematic observation and measurement of social signals and actions that animals use to attract, assess, and engage potential mates, providing a window into motivated behavior and neural control. Researchers typically quantify behaviors such as orientation, approach, vocalization, display, pursuit, and receptivity under defined social and environmental conditions, then compare their timing, frequency, and sequence. In neuroscience, these measurements help relate behavioral changes to brain circuits, sensory processing, hormones, and neuromodulators involved in social decision-making. The approach supports studies of sexual motivation, communication, learning, and the behavioral effects of neural manipulation or disease.

2017

This protocol describes a Drosophila learning and memory assay called courtship conditioning. This classic assay is based on a reduction of male courtship behavior after sexual rejection by a non-receptive premated female. This natural form of behavioral plasticity can be used to test learning, short-term memory, and long-term memory.

Drosophila Courtship Conditioning: A Method to Test Learning and Memory In Flies

2023

This video describes a classical conditioning assay that tests learning and memory in Drosophila, called courtship conditioning. The assay is based on the reduction of male courtship after experiencing a rejection by a non-receptive premated female. The example protocol shows a setup for the procedure that can be used to assess short- and long-term memory.

2016

An efficient and simple methodology for computer-based analysis of nematode swimming behavior in liquid is described. The method requires little to no investment for C. elegans laboratories. The hardware used is standard, and the computer software for the behavioral analysis (CeleST) is an open source one.
