Ethanol Extraction

Ethanol extraction is a solvent-based method that separates small molecules from biological materials, making it useful for preparing samples for chemical and biochemical analysis. In neuroscience, tissue or fluid is mixed with ethanol, which dissolves many polar and moderately nonpolar compounds while precipitating proteins and disrupting cellular structures; centrifugation then separates the extract from insoluble material. The resulting fraction can be concentrated and analyzed for neurotransmitters, metabolites, lipids, or other neurochemical components, depending on the extraction conditions and analytical method. This approach supports studies of brain chemistry, cellular signaling, disease-related biochemical changes, and the effects of drugs or environmental exposures.

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Biofuels: Producing Ethanol from Cellulosic Material

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University In this experiment, cellulosic material (such as corn stalks, leaves, grasses, etc.) will be used as a feedstock for the production of ethanol. The cellulosic material is first pretreated (ground and heated), digested with enzymes, and then fermented with yeast. Ethanol production is monitored using an ethanol probe. The experiment can be extended to optimize ethanol production by varying the feedstock used,...

Source: Bucher, T., et al. Methodologies for Studying B. subtilis Biofilms as a Model for Characterizing Small Molecule Biofilm Inhibitors. J. Vis. Exp. (2016).This video demonstrates how biofilm matrix integrity influences bacterial resistance to ethanol by comparing colony-forming units in untreated and inhibitor-disrupted biofilms. Reduced CFUs in inhibitor-treated samples demonstrate that the extracellular matrix is essential for protecting biofilm-embedded cells from antimicrobial stress.
