Ex Vivo Corneal Model

An ex vivo corneal model is an isolated cornea maintained outside the organism to preserve native tissue structure for controlled biological study. In organ culture, the cornea remains viable under defined conditions, allowing researchers to manipulate injury, inflammation, or pharmacological exposure while measuring epithelial repair and interactions between corneal cells and sensory nerves. In neuroscience, this model supports investigation of corneal innervation, trigeminal nerve signaling, ocular pain, and neuroimmune communication. It also provides a controlled platform for testing therapies that influence nerve regeneration or sensory function, helping bridge cellular experiments and in vivo studies while reducing experimental complexity.

In this video article we describe the use of a new ex vivo model of acute herpes simplex virus type I corneal epithelial infection.

Porcine Ex vivo Cornea Model of Bacterial Keratitis: A Technique to Establish Bacterial Infection in Corneal Epithelial Cells

2025

This video describes a protocol to set up an ex vivo model for bacterial keratitis using a corneoscleral button dissected from the porcine eye. Inoculated bacteria traverse the corneal epithelial incisions to invade the underlying stroma and establish an infection within the corneal tissues that mimic the in vivo infection.

Laser-assisted Selective Ablation of Porcine Corneal Endothelial Cells: A Technique to Induce Focused Porcine Corneal Endothelial Injury for Ex Vivo Studies
