Flight Muscle Injection

Flight muscle injection is a targeted laboratory technique that introduces a measured substance directly into the muscles insects use for wing movement. It matters in neuroscience because it links muscle activity with neural control of locomotion. Using a fine needle or micropipette, researchers penetrate the muscle and deposit a test solution at a defined site and dose; localized delivery can alter muscle physiology or mark nearby structures for later analysis. This approach supports studies of motor circuits, neuromuscular transmission, muscle function, and flight behavior, helping clarify how neural signals generate coordinated movement and how disruptions affect motor performance.

Drosophila is a powerful model to understand fundamental mechanisms of myogenesis. This protocol for the dissection and preparation of thorax hemi-sections enables microscopy analysis of indirect flight muscle from both pupal and adult stages. This protocol enables confocal imaging of cellular morphology, protein localization, muscle structure, and multiple other aspects of myogenesis.

A Simple Flight Mill for the Study of Tethered Flight in Insects

Flight in insects is influenced by a number of factors and the propensity to disperse is an important variable in understanding insect ecology and biological control strategies. We describe the construction and use of a simple, relatively inexpensive, and flexible flight mill for measuring parameters of tethered flight in insects.

We report a method to quantify muscle area, which is an indirect method to determine muscle mass in Drosophila adults. We demonstrate the application of our methodology by analyzing the indirect flight muscles in a Drosophila model of Myotonic Dystrophy disease.

Drosophila flight muscle is a powerful model to study transcriptional regulation, alternative splicing, metabolism, and mechanobiology. We present a protocol for dissection of fluorescent-labeled flight muscle from live pupae to generate highly enriched samples ideal for proteomics and deep-sequencing. These samples can offer important mechanistic insights into diverse aspects of muscle development.
