Hyperstack Imaging

Hyperstack imaging is a microscopy data format and analysis approach that organizes multidimensional image series across spatial depth, time, and fluorescence channels, allowing complex biological events to be examined in one dataset. By linking x-y image planes into z-stacks and combining them with repeated time points or multiple labels, researchers can reconstruct three-dimensional structures and track changes while preserving relationships among channels and measurements. In neuroscience, this approach supports analysis of neuronal morphology, synaptic organization, activity-linked signals, and interactions among cells in intact tissue or culture. It strengthens visualization and quantitative comparison of dynamic neural processes.

Here, we present a method for investigating neurite morphogenesis in postnatal mouse retinal explants by time-lapse confocal microscopy. We describe an approach for sparse labeling and acquisition of retinal cell types and their fine processes using recombinant adeno-associated virus vectors that express membrane-targeted fluorescent proteins in a Cre-dependent manner.
