Immunostaining Brain Cells

Immunostaining brain cells is a laboratory method that uses antibodies to identify and visualize specific proteins in neural tissue, helping researchers determine cell identity, structure, and function. The technique relies on selective antigen–antibody binding: a primary antibody recognizes a target protein, while a labeled secondary antibody produces a fluorescent or enzymatic signal that can be detected by microscopy. In neuroscience, immunostaining distinguishes neurons, astrocytes, microglia, and other cell types, and reveals the distribution of molecules within cells and brain regions. These patterns support studies of neural development, connectivity, injury, neurodegenerative disease, and responses to experimental treatments.

This video demonstrates a technique for immunostaining cells encapsulated in a 3D hydrogel. The hydrogel culture containing neural progenitor cells is fixed, permeabilized, and treated with a blocking solution to prevent non-specific immunostaining. A primary antibody cocktail is added to bind specific proteins, followed by fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted on a slide and observed under a confocal microscope.
