Interleukin-6 Microinjection

Interleukin-6 microinjection is an experimental technique that delivers the cytokine interleukin-6 directly into a selected brain region to examine its localized effects on neural function. Using a fine micropipette, researchers administer a controlled volume and concentration, allowing IL-6 to bind cellular receptors and activate signaling pathways such as JAK/STAT3 in nearby cells. In neuroscience, this approach helps investigate how neuroinflammation influences neuronal activity, glial responses, behavior, and disease-related processes. By controlling injection site and timing, researchers can distinguish regional IL-6 actions and evaluate its potential contribution to conditions involving inflammation and altered brain function.

To model how inflammatory signals influence neurodevelopment in genetically susceptible organisms, we present a protocol for microinjecting IL-6 into the duct of Cuvier (common cardinal vein) of zebrafish larvae, enabling controlled induction of inflammation and assessment of its developmental impact in the context of tuberous sclerosis complex (TSC).
