Micro-drill Holes

Micro-drill holes are precisely made, small openings in bone, commonly the skull, that provide controlled access to underlying nervous tissue while limiting disruption to surrounding structures. In neuroscience, a fine drill removes a localized area of cranial bone, often under stereotaxic guidance, so researchers can position electrodes, inject substances, or access the brain for imaging and other procedures. Hole size, location, drilling speed, and cooling help balance access with tissue preservation and reduce mechanical or thermal injury. This approach supports experiments involving neural recording, stimulation, drug delivery, and repeated observation, making it useful for linking brain structure and activity to behavior and disease.

Intervertebral disc degeneration is a significant contributor to back pain and a leading cause of disability worldwide. Numerous animal models of intervertebral disc degeneration exist. We demonstrate an ovine model of intervertebral disc degeneration, utilizing a drill bit, which achieves a consistent disc injury and reproducible level of disc degeneration.

This protocol describes performing a craniectomy using a high-speed pneumatic drill on a 3-month-old Danish Landrace pig. The access is made through the frontal bone and reveals the ventral dura mater and underlying cerebral hemispheres. This procedure allows for access to a large portion of the pig brain.

Recording electrodes are placed in the olfactory bulb, reference electrodes in the frontal cortex, and micro-drive array electrodes in the hippocampus.
