Mitochondrial Stress Assay

A mitochondrial stress assay is a laboratory method used to evaluate how mitochondria respond to conditions that challenge cellular energy production and survival, making it valuable for studying neuronal health. These assays quantify indicators such as oxygen consumption, extracellular acidification, ATP production, membrane potential, or reactive oxygen species before and after metabolic perturbation. In neuroscience, they help reveal impaired bioenergetics, oxidative stress, and reduced respiratory capacity in neurons and glial cells. The results support investigation of neurodegenerative disease mechanisms, effects of toxic or therapeutic compounds, and cellular responses to injury, while helping identify mitochondrial pathways that may be targeted to preserve neural function.

Comet Assay as an Indirect Measure of Systemic Oxidative Stress

Comet assay measures DNA breaks, induced by different factors. If all factors (except oxidative stress) causing DNA damage are kept constant, the amount of DNA damage is a good indirect parameter of oxidative stress. The goal of this protocol is to use comet assay for indirect measurement of oxidative stress.
