Parallel Fiber Stimulation

Parallel fiber stimulation is an experimental neuroscience technique that activates the axons of cerebellar granule cells to examine how distributed inputs influence neural circuits. Electrical stimulation recruits parallel fibers, which release glutamate onto Purkinje cell dendrites and molecular-layer interneurons; when paired with climbing fiber activity, the resulting calcium signals can induce long-term depression at parallel fiber–Purkinje cell synapses. Researchers use this approach in cerebellar slices and living preparations to investigate synaptic integration, activity-dependent plasticity, and the cellular mechanisms underlying motor learning. Controlled stimulation also helps relate circuit-level responses to the timing and strength of sensory and motor signals.

The development of optogenetics now provides the means to precisely stimulate genetically defined neurons and circuits, both in vitro and in vivo. Here we describe the assembly and implantation of a fiber optic for chronic photostimulation of brain tissue.

In Vivo Single-Fiber Recording of Sciatic Nerve C-Fibers in Rats

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.

Dorsal Column Steerability with Dual Parallel Leads using Dedicated Power Sources: A Computational Model

Using a mathematical model of spinal cord stimulation, we found that a multi-source system with independent power sources for each contact can target more central points of stimulation on the dorsal column (100 vs 3) and has 50-fold more field steering resolution (0.02mm vs 1mm) than a single-source system.
