Receptor Labeling

Receptor labeling is a method for identifying and visualizing specific cell-surface or intracellular receptors, helping researchers map how cells detect and respond to signals. It uses a selective ligand, antibody, or engineered binding molecule attached to a detectable tag, such as a fluorescent or radioactive marker; binding localizes the receptor and can reveal its abundance or distribution. In neuroscience, receptor labeling helps characterize neurotransmitter and neuromodulator systems, distinguish receptor populations, and examine changes associated with development, learning, disease, or drug exposure. These measurements connect receptor location and density with neural circuit function and pharmacological responses.

Guanosine triphosphate (GTP) binding is one of the earliest events in G-Protein-Coupled Receptor (GPCR) activation. This protocol describes how to pharmacologically characterize specific GPCR-ligand interactions by monitoring the binding of the radio-labeled GTP analog, [35S]guanosine-5'-O-(3-thio)triphosphate ([35S]GTPγS), in response to a ligand of interest.

Use of Label-free Optical Biosensors to Detect Modulation of Potassium Channels by G-protein Coupled Receptors

Optical biosensor techniques can detect changes in mass near the plasma membrane in living cells and allow one to follow cellular responses in both individual cells and populations of cells. This protocol will describe detection of the modulation of potassium channels by G-protein coupled receptors in intact cells using this approach.

Following the preparation of a 64Cu-modified monoclonal antibody binding to a transgenic murine T cell receptor, T cells are radiolabeled in vivo, analyzed for viability, functionality, labeling stability and apoptosis, and adoptively transferred into mice with an airway delayed-type hypersensitivity reaction for non-invasive imaging by positron emission tomography/computed tomography (PET/CT).
